FokI是一种存在于细菌Flavobacterium okeanokoites的type IIS限制酶，含有位于N端的DNA结合区块（N-terminal DNA-binding domain），以及一个位于C端的非专一性DNA切割区块[2]。当此酵素与DNA辨识并结合于5'-GGATG-3'识别位点时，将会把位于结合位置下游的9到13个核苷酸切除（不需特定序列）[3]。FokI的分子量为65.4 kDA，含有587个氨基酸。
