Katepsin K (EC 3.4.22.38 ili katepsin O2 je katepsinski enzim iz porodice cistein peptidaza.[5][6][7][8][9][10][11][12]

Katepsin K katalizira hemijsku reakciju u kojoj se odvija:

široka proteolitska aktivnost na malim molekulama supstrata. Glavna odrednica specifičnosti je P2, koja je prvenstveno na pozicijama leucina metionina > fenilalanina, uz odsustvo arginina

Ovaj oblik katepsina, kod sisara se prominentno ispoljava u osteoklastima.
