Barvanje s hematoksilinom in eozinom

Barvanje s hematoksilinom in eozinom ali okrajšano barvanje HE[1] je eden od osnovnih postopkov barvanja tkiv v histologiji.[2][3][4] V diagnostiki predstavlja najpogosteje uporabljen postopek barvanja[2] in pogosto predstavlja zlati standard.[5] Gre za zaporedno barvanje bazofilnih struktur s hematoksilinom in acidofilnih struktur z eozinom.[1] Hematoksilin obarva celično jedro vijoličastomodro, eozin pa medceličnino in citoplazmo rožnato; ostale celične strukture se obarvajo z različnimi odtenki in kombinacijami teh dveh barv.[6][7] Patologi lahko tako zlahka prepoznajo celično jedro in citoplazmo in nasploh zgradbo tkiva v preučevanem vzorcu.[8] Barvanje s hematoksilinom in eozinom je leta 1876 prvi uvedel A. Wissowzky.[9][8]

Uporaba

Barvanje s hematoksilinom in eozinom je eden od osnovnih postopkov barvanja v histologiji,[4][8][3][6] predvsem zato, ker se lahko opravi hitro,[8] ni drago, obarva in naredi prepoznavne številne celične strukture[10][11][8][6][5] ter se lahko uporablja za diagnostiko številnih histopatoloških stanj.[9] Rezultat barvanja HE tudi ni zelo odvisen od snovi, uporabljenih za fiksacijo vzorca tkiva, ali manjših odstopanj oziroma razlik v protokolih barvanja.[12] Vendar pa ne omogoča razpoznave vseh tkiv, celičnih struktur ari razporeditve kemijskih snovi v celicah,[10] za te namene so potrebna specifičnejša barvila in postopki barvanja.[11][8]
